DMSO Grades, Purity, and Regulatory Assurance: Understanding Quality Differentiation and Manufacturing Traceability

Widely used in the pharmaceutical industry for drug discovery, the synthesis of Active Pharmaceutical Ingredients (APIs), and analytical applications, Dimethyl Sulfoxide (DMSO) is a powerful solvent known for its ability to quickly penetrate skin and biological membranes. It’s also an excipient solvent in pharmaceutical drug delivery systems.

The exact purity of DMSO dictates whether it’s suitable and safe across different applications. DMSO is manufactured and classified into specific grades based on its intended use. As such, high-purity DMSO must meet strict regulatory requirements.

Contract Pharma: How does gChem ensure products consistently meet the necessary specifications for their intended use?

Sandra Reid: At gChem we manufacture multiple grades of Dimethyl Sulfoxide in different manufacturing units, intended for different applications. What does not vary is our commitment to quality.  We are dedicated to satisfying customer needs with high quality sustainable products and services for diverse markets around the world. For example, applications in life science market vary from the highest purity, sterile DMSO used in injectables (PROCIPIENT® Sterile) to high quality DMSO used for Active Pharmaceutical Ingredient (API) synthesis where the DMSO does not remain in the final API. Therefore, the purity, compliance, complexity, and manufacture of these varying grades, and applications in between, are different.

Product specifications vary between grades. Some examples are assay, water content, or UV absorbance. The specification is determined by the governing quality certification or registration of the grade of Dimethyl Sulfoxide. The Quality Management System maintains requirements for GMP manufacturing and ISO 9001 certification deemed essential for quality effectiveness. It is our goal to achieve 100% product delivery in-specification material, regardless of the specification. gChem follows an internal procedure securing customer approval of the product specification needed for their application. For every customer order, products undergo a thorough testing to be approved and released by the quality department. Products found to be out of spec are quarantined and managed with appropriate disposition procedures.

Product specifications for our PROCIPIENT® dimethyl sulfoxide are governed by its compliance to the USP and PhEur Monograph. This grade of DMSO is manufactured under cGMP conditions, in compliance with ICH Q7 guidelines. In contrast, for life science analytical applications we frequently recommend DMSO ACS Reagent Grade which maintains a tight specification governed by our own quality system, ISO 9001 certification. Contract Pharma: How does gChem ensure traceability within the excipient supply chain?

Sandra Reid: This is a great question. Excipients in respective countries have different standards but in no country is it required that excipients have the same requirement as an Active Pharmaceutical Ingredients. At gChem, we take the responsibility of safety, purity, and fitness for use as an excipient to the highest standard. Specifically for excipient use, we recommend PROCIPIENT®. Because we manufacture PROCIPIENT® under current Good Manufacturing Practices (cGMP) guidelines and we maintain a Type II Drug Master File (DMF) on file with the FDA, and DMF at Health Canada, and a CEP granted by EDQM, the product itself is an API. PROCIPIENT® provides audit, traceability and inspection history which is maintained by our quality department. 

Contract Pharma: How does gChem foster partnerships with pharmaceutical developers to support drug development and enhance the quality of pharmaceutical products?

Sandra Reid: For all customers, enhancing safety specifically is an easy choice! If a formulator needs an aprotic solvent, DMSO is an obvious choice. We have data supporting the safety of DMSO compared with other solvent options. DMSO is not carcinogenic. DMSO is not mutagenic. And DMSO is not a reproductive toxin. DMSO is a naturally occurring substance, we just happen to manufacture it on purpose for its wide utility across many industries.

The quality of pharmaceuticals is always improving but what I see rapidly changing is the quality of the experience for the patient. For example, historical chemotherapy treatments involved intravenous administration that could take hours typically in a controlled environment. While I know this is still a necessary treatment for some, patients now have the option of immunotherapy subcutaneous injections which may take 5 minutes at a doctor’s office or even home administration. To me, this is enhancing quality of pharmaceuticals!  A patient being able to self-administer a SC injection which may be possible because of PROCIPIENT® enhances our industry. Using a topical patch which delivers a necessary API because of PROCIPIENT® enhances our industry!

Contract Pharma: When sourcing gChem DSMO what indications, labels, paperwork, or certificates should a buyer request ensuring a high-quality product?

Sandra Reid: This is a great question and so critical for the pharmaceutical formulator to ensure they are sourcing a high-quality DMSO. There are few documents that are helpful to determine the source and manufacturing condition of the DMSO. Upon request, gChem will provide a COO which is a Certificate of Origin. This indicates the origin of the product. gChem is the only US manufacturer of DMSO to our knowledge. gChem’s manufacturing facility is audited by the FDA and we have a registered site which can be found on the FDA’s website Drug Establishments Current Registration Site. Gaylord Chemical LLC is a registered site and with proper internal approval we will share a statement regarding the latest FDA audit, date, and status. If a verified customer is filing an IND, NDA, or ANDA, gChem will share a Letter of Authorization (LOA) allowing the customer to reference product with their filing. gChem also has updated ISO certification for various grades of DMSO and this certification is shared with customers upon request. These are a few important differentiators that can help the client determine the manufacturing source, condition of which the DMSO is manufactured under, and the relevant status maintained.
